PRODUCTION OF NANO PARTICLE CHITOSAN FROM CRAB SHELL AS BLOOD COAGULATION AGENT

Chitosan is a chitin derivative natural polymer compound isolated from fishery waste, such as crab shell which is the most source of chitosan. Nanochitosan as a drug delivery offers delivery with harmless routes such as oral, nose, eye, and absorbency. Nanochitosan can be absorbed by human organs which have the ability to penetrate nano-sized membranes such as the kidneys, liver, and lungs where nanoparticles measuring 1-1,000 nm. Chitosan nanoparticles were prepared by ionic gelation techniques. The mechanism of this method is the formation of chitosan nanoparticles based on the electrostatic interaction between the positive amine group on chitosan (-NH2) and the negative charge group from tripolyphospate polyanion (TPP). The initial stage is the manufacture of chitosan, crushed crab shell waste to the size of 60 mesh, after being smoothed then deproteination to eliminate the protein content by dissolving in NaOH 3 N, then crude chitin demineralized to eliminate the mineral content contained in the crab shell with the addition of 1 N HCl and heating and stirring. The chitin obtained then goes into the deacetylation stage to remove the acetyl group in the crab shell by adding 50% NaOH and is done by heating and stirring. The chitosan obtained was then dissolved in 3% lactic acid to obtain sol gel phase chitosan, then NaTPP was added to reduce the particle size of chitosan to nano size. The crab obtained nanocitosan has a distribution size of 2822.5 nm with variations in the addition of NaTPP concentration of 0.1%. The resulting nanochitosan was tested to increase the speed of blood clotting which was proven to shorten the blood clotting time from 240 seconds to 30 seconds and was able to inhibit the activity of Staphylococus aureus bacterial with the addition of nanopropolis.
